The present invention relates to a method of diagnosing bovine mastitis using a primer for detecting a haptoglobin gene, and a real-time polymerase chain reaction (PCR) kit for the method. Specifically, the present invention relates to a method of diagnosing mastitis of milk-producing milk cows by using a milk sample or DNA separated from the milk sample as a template, and performing a real-time PCR using a primer set for detecting a haptoglobin gene; and to a real-time PCR kit for easily performing the diagnosis method. A method of diagnosing bovine mastitis using a primer for detecting a haptoglobin gene according to the present invention is capable of performing the diagnosis of bovine mastitis and the quality analysis of milk very effectively even in cases of other causes as well as cases due to microbial infections, by promptly and accurately checking the concentration of somatic cells having the haptoglobin gene from the sample in a simple manner. Particularly, the present invention is capable of performing diagnosis or analysis more accurately by combining the method of diagnosing bovine mastitis with PCR on microorganisms causing bovine mastitis. The method of the present invention can be used very usefully in general livestock farms since the method has merits that it is performed very simply, promptly, and accurately. Accordingly, the method of the present invention is capable of preventing, in advance, a problem that an overall quality of milk is lowered due to additional infection or milk mixing since the method is capable of quickly dealing with the problem by isolating milk cows having bovine mastitis when bovine mastitis occurs.
