Provided are a composition for enhancing immunity, containing a milk protein with a high content of organic selenium as an active ingredient, and a method for preparing the same. The milk protein with a high content of organic selenium prepared by mixing mixed powder, which is prepared by mixing whey powder and selenium power, with water and then heating the same, increases the bioavailability of minerals. In addition, the composition has immune enhancing activities by: increasing the number of immune cells such as macrophages, neutrophils, monocytes, etc., which primarily defend against pathogenic microorganisms, and natural killer (NK) cells in the blood; increasing the number of CD4 positive helper T cells (T_H cells) and CD8 positive cytotoxic T cells (T_C cells) in lymph nodes; and increasing the secretion of tumor necrosis factor- (TNF-) and interleukin-1 (IL-1), the immune-related cytokines secreted from activated macrophages, when the composition is orally administrated, thereby being able to be used as an active ingredient of a food composition or a pharmaceutical composition for enhancing immunity.
