The present invention relates to a method for distinguishing between Dioscorea nipponica Makino and Dioscorea quinquelobata Thunb., comprising: (a) a step of dyeing internal tissues of samples that are suspended to be Dioscorea nipponica Makino or Dioscorea quinquelobata Thunb., or measuring the content of dioscin; and (b) a step of distinguishing the dyed tissues in which a periderm is dyed red, and which is consisted of phellem only as Dioscorea quinquelobata Thunb., distinguishing the dyed tissues in which a periderm is dyed blue, and which is consisted of phellem, phellogen, and phelloderm as Dioscorea nipponica Makino, or distinguishing the dyed tissues as Dioscorea nipponica Makino or Dioscorea quinquelobata Thunb. according to the content (mg) of dioscin measured based on dry weights (g) of the samples. Although a derived plant of Dioscoreae Nipponicae Rhizoma is Dioscorea nipponica Makino, Dioscorea quinquelobata Thunb., which is an allied species, is currently also distributed as Dioscoreae Nipponicae Rhizoma without distinction. As disclosed in Figs. 4 and 5, appearances of Dioscorea nipponica Makino or Dioscorea quinquelobata Thunb. are almost similar to each other. Particularly, since Dioscoreae Nipponicae Rhizoma is circulated after cutting and drying, it is very difficult to distinguish the cut and dried Dioscoreae Nipponicae Rhizoma with the unaided eye (Fig. 6). Therefore, the present invention has suggested a method for distinguishing between Dioscorea nipponica Makino and Dioscorea quinquelobata Thunb. easily and promptly by a high speed liquid chromatography method or a double staining method using safranin and a fast green staining reagent as a method for distinguishing Dioscorea nipponica Makino and Dioscorea quinquelobata Thunb., Dioscoreae Nipponicae Rhizoma medicinal ingredients which are difficult to be distinguished with the unaided eye in the distribution process.;COPYRIGHT KIPO 2016
