The present invention relates to a defect verification system of a spent nuclear fuel assembly using a gamma-ray imaging device and a verification method thereof, and specifically, to a defect verification system of a spent nuclear fuel assembly using a gamma-ray imaging device for verifying whether the spent nuclear fuel assembly is defective or not. The defect verifying system comprises: a Compton image acquiring portion (100) which includes a plurality of gamma-ray detectors, which measure a reaction position of a gamma-ray, produced from spent nuclear fuel, and emitted gamma-ray energy, and acquires a tomographic Compton image about the spent nuclear fuel assembly using the measured reaction position of the gamma-ray and the gamma-ray energy; and an integrated management portion (200) which receives the tomographic Compton image from the Compton image acquiring portion (100), and determines an availability of a partial defect or a bias defect of the spent nuclear fuel.
