The present invention relates to a method of operating a sedimentary basin. The exploitation of the sedimentary basin is allowed by the choice of zones of interest (ZI) within the basin, zones (ZI) being determined by means of maps (CAR) of organic carbon content TOC and index of hydrogen IH of at least one sedimentary layer. According to the invention, the TOC organic carbon content (CAR) and hydrogen index 1H maps are obtained taking into account current values of TOC A (P) and IH A (P) at the well and taking into account the CB bathymetry maps and CS sedimentation velocity.
