PROBLEM TO BE SOLVED: To provide methods of selectively amplifying an Internal Transcribed Spacer region (ITS region) which is present between rRNA genes of a plant symbiotic filamentous fungus, the methods being used in a diversity analysis of plant symbiotic filamentous fungus.SOLUTION: By performing PCR with a reaction solution which contains a DNA derived form a plant sample, a primer set (at least either one primer contains a locked nucleic acid (LNA)) consisting of two primers targeting an ITS region of a plant symbiotic filamentous fungus, and an LNA-containing oligonucleotide corresponding to DNA of a plant designed in a position competing with the primer of the primer set, the LNA-containing oligonucleotide inhibits amplification of the ITS region of a plant and selectively amplifies the ITS region of plant symbiotic filamentous fungus with the primer.SELECTED DRAWING: Figure 1
