Preparation of recombinant protein, useful for e.g. blood factors, using serum- and protein-free medium supplemented with specific amino acids for increased productivity

Preparation, especially on a large scale, of recombinant products (A) using a medium that is free of serum and proteins but contains at least one of the L-amino acids (aa) Asn, Cys, (Cys)2, Pro, Trp or Gln, is new. Independent claims are also included for: (1) an isolated, stable, recombinant cell clone, originally produced by culturing a source clone on serum-containing medium, is adapted to serum- and protein-free medium and tested for stable production of (A) and a selected colony cloned under serum- and protein-free conditions (this clone is replicated under similar conditions to produce a culture, in a bioreactor, of stable cells and protein is harvested from the culture supernatant by the method of AT 407255); and (2) an isolated, stable recombinant cell clone produced this way.
