A method of in vitro generation of an isolated cell population comprising anti-third-party cells that do not induce graft versus host disease (GVHD) that have a central memory T lymphocyte (Tcm) phenotype, said T-cell phenotype comprising central memory (Tcm) a CD8 + / CD62L + / CD45RO + badge and in which at least 50% of the isolated cell population have such a badge, said cells being cells that induce tolerance and are able to lodge in the lymph nodes after transplantation , the method comprising: (a) contacting peripheral blood mononuclear cells (PBMC) with a third-party antigen or antigens in the presence of IL-21 under conditions that allow the removal of GVH reactive cells and (b) culturing said cells resulting from step (a) in the presence of IL-15 in an antigen-free environment under conditions that allow proliferation of cells comprising said pheno type of central memory T lymphocyte (Tcm), thereby generating the isolated cell population.
