The present invention provides a method for developing a photo-thermo-sensitive sterile line by means of CalS5 gene mutation, which is implemented by reducing the expression of callose synthase associated with pollen growth in a plant. In the present invention, by adjusting the expression or the activity of CalS5 protein associated with pollen growth in a plant, the fertility of the plant is adjusted, and accordingly conversion between sterility and fertileness can be implemented, and a breeding method for a plant sterile line is simplified.
