METHOD FOR PREPARING MEMBRANE VESICLES SUITABLE FOR BIOPHYSICAL ANALYSIS

Provided is a method for preparing a membrane vesicle population, the method including: (1) a step for ultrafiltration treatment of a sample solution including membrane vesicles; (2) a step for substituting the sample solution obtained from step (1) by a colloidal flocculant-free buffer solution, and thereby preparing a buffer solution including membrane vesicles; and (3) a step for separating the membrane vesicles included in the buffer solution obtained from step (2) by density gradient centrifugation. Also provided is a membrane vesicle population separated and purified from a sample solution, wherein: the membrane vesicles included in the membrane vesicle population are dispersed; and (i) the membrane vesicles included in the membrane vesicle population maintain a surface charge when present in the sample solution prior to separation and purification, (ii) the membrane vesicles included in the membrane vesicle population maintain their form when present in the sample solution prior to separation and purification, and/or (iii) the membrane vesicle population maintains a grain size distribution pattern when present in the sample solution prior to separation and purification.
