The present invention relates to a method for expressing water solubility of codon-optimized transmembrane receptor activator of nuclear factor (NF)-kB ligand (mRANKL) in E. coli, and to optimization of expression through a response-surface analysis scheme. In particular, provided is a codon-optimized RANKL gene represented by SEQ ID NO: 1. Moreover, provided are a recombinant expression vector including the codon-optimized RANKL gene, and a recombinant microorganism transformed by the recombinant expression vector. Furthermore, provided is a method for producing an RANKL protein, wherein the method comprises a step of culturing the recombinant microorganism. In addition, the inventors have continuously induced physiological numbers of M cells and immunity cells by administrating mRANKL through a penetration scheme, and have suggested a new strategy for improving efficiency of oral vaccines by continuously transferring M cell-targeted antigens through a mucosal adhesive and pH sensitive oral delivery system.;COPYRIGHT KIPO 2016
