The present invention relates to a calibration method and system of an electrochemical sensor detecting a biomaterial which is a detection target through current measurement and, more specifically, relates to a calibration method and system of an electrochemical sensor using a fluorescence absorption phenomenon configured to make a sensing value a reference point in a state where a target material is not coupled to a sensor and fluorescence does not diverge; and to calculate an amount of the target material in accordance with how much the sensing value is changed based on the reference point in a state where the target material is coupled to the sensor and the fluorescence diverges to be able to improve sensing accuracy and also check the amount of the target material in the electrochemical manner, as well as being able to visually quickly check a presence of the target material.
