The present invention relates to a device and a method for generating a distance measuring equipment (DME) pulse used in a navigation system. Especially, a device and a method for generating a DME pulse used in a navigation system generate a plurality of DME pulses by using a gene algorithm by inputting one basic DME pulse, filter in three steps by calculating a power and a shape of a spectrum and a distance measurement accuracy with respect to the generated DME pulse and comparing with a DME pule standard of an international civil aviation organization (ICAO) and a set value, and generate a final DME pulse with high distance measurement accuracy by repeatedly performing the above processes until one DME pulse is selected. The device for generating a DME pulse used in a navigation system comprises: a pulse standard storage part; a control part; a pulse spectrum power calculation part; a pulse shape calculation part; and a distance measurement accuracy calculation part.
