The invention is a method for observing a sample comprising illuminating the sample with a light source and acquiring an image of the sample by an image sensor, the sample being disposed between the image sensor and the light source. The acquired image, also referred to as the hologram, is the subject of the application of iterative steps, so as to estimate a complex image of the sample in a reconstruction plane or in a detection plane, according to which extends the image sensor. This complex image can allow a characterization of the sample.
