Sample loading buffer for visualisation of target molecules in polyacrylamide gel electrophoresis

A sample loading buffer for visualisation of protein molecules during electrophoresis of a sample containing the molecules and loaded together with the buffer into a well in a polyacrylamide gel plate comprises an activated aromatic dye, such as Remazol, an alkaline buffer solution, such as sodium carbonate or bicarbonate and a density-increasing agent such as sorbitol, which is inert in relation to the dye and the buffer solution and which increases the density of the buffer solution to promote subsidence in the well. The dye, but not the buffer solution, contains a reactive group producing covalent attachment to the protein molecules in the sample so that during electrophoresis producing migration of the molecules across the gel plate the dye-marked molecules become visible to the user.
