PROBLEM TO BE SOLVED: To provide a more robust and efficient split intein for use in refinement of various proteins and application of modifications to proteins.SOLUTION: The method of the present invention involves a fusion protein comprising a split intein N-terminal fragment and a polypeptide in which the N-terminal fragment comprises a sequence selected from 19 specific amino acid sequences or variants thereof. The polypeptide may be an antibody or a fragment thereof. A split intein C-terminal comprises a specific sequence or a variant thereof. An intein intermediate body is formed by the fusion protein comprising the split intein N-terminal fragment binding with the split intein C-terminal fragment and a protein nucleophilic reagent complex is formed by bringing a nucleophilic reagent into contact with the intein intermediate body.SELECTED DRAWING: None
