The present invention relates to a method of concentrating a biological sample containing nucleic acids by using magnetic chitosan microparticles and then performing a PCR reaction on the nucleic acid captured on the microparticles. Chitosan microparticles added to a PCR compatible pH biological sample are mechanically agitated to provide cell lysis and simultaneous DNA capture and then serve as a solid support for the nucleic acid template during the PCR reaction. Since chitosan microparticles are utilized for lysis and it is not necessary to remove nucleic acids from the microparticles prior to PCR, the ease of sample preparation procedures is dramatically improved.
