A method of making a live cell construct comprises: (a) providing a non-cellular support having a top surface and a bottom surface; (b) contacting live undifferentiated cells with a non-cellular support; (C) growing a gastrointestinal epithelial cell monolayer on the top surface. In some embodiments, the living cells in the monolayer are: (i) undifferentiated cells (eg, stem cells or progenitor cells); and (ii) optionally, but in some embodiments preferably differentiated Cells (eg, enterocytes, panate cells, enteroendocrine cells, tufted cells, microcells, intraepithelial lymphocytes and / or goblet cells). Also described are constructs produced by such methods and methods of using them (eg, in high-throughput screening).
