The present invention relates to the design of two sequences optimized for prokaryotes and eukaryotes for the synthesis of a recombinant protein comprising two Entamoeba Histolytica proteins: 1) the LECAg fragment (sequence encoding this region is between up 758aa to 1134aa of the 170kDa lectin); and 2) EhCP (a conserved region of the cysteine ??proteases representing a fragment of up to 111aa). The present invention further relates to the use of Pichia pastoris expression vectors for synthesizing such proteins from the expression vectors also containing an identification system, purification, expression of E. histolytica proteins, immunogenicity and a site for enhancing endogenous class I. Said sites are ordered as follows (sequence 6xHis, cleavage sites for enterokinase, PEDIII, LECAg fragment or EhCP, KDEL3); the proteins obtained can be used as vaccines against E. Histolytica or to generate antisera. On the other hand, the proteins are used in clinicians diagnostic methods by sero logy for the identification of antibodies produced against E. Histolytica.
