A bio-photon spectrum detection system and detection method. The bio-photon spectrum detection system is formed by adding a beam-splitting sub-system composed of a slit (3) and a grating (2) based on an ultra-weak bio-photon imaging sub-system. The grating (2) is an optical element composed of a large number of parallel slits having equal widths and equally spaced, and disperses light by utilizing the principle of multiple-slit diffraction. According to the principle of multiple-slit diffraction, the ultra-weak bio-photons emitted by a sample to be detected pass through the slit (3) and form a line light source; and the line light source radiates the grating and forms bright fringes arranged in an equally spaced mode after multiple-slit diffraction, and the distance between the bright fringes is related the wavelength of the bio-photons. The imaging sub-system (1) is used for image recording and analysis. Experimental detection has proved that the bio-photon spectrum detection system has high sensitivity and can detect the spectrum of ultra-weak bio-luminescence, and also has the advantages of being simple and convenient to operate, having good extensibility, abundant functions, etc., and can be widely applied to the field of life science researches, medical clinical examinations, agricultural production, food security, environment protection, etc.
