The present invention relates to a system and a method of measuring three-dimensional displacement of a structure using a single camera. The system of measuring three-dimensional displacement of a structure using a single camera comprises: an image acquisition unit for acquiring sequential images of a structure through a single camera; an interest area selection unit for selecting an interest area of a structure of which desirable displacement to be extracted from the image, and extracting the same; a coordinate calculation unit for extracting a coordinate change and a size change of a target from the image extracted from the interest area selection unit; and a calibration unit for extracting three-dimensional displacement of the structure from the extracted coordinate and size changes. According to the present invention, the coordinate change and the size change of the target are calculated from the image of the structure photographed by a single camera, and thus high reliability is ensured.
