The present invention provides a method for regulating the expression of an endogenous cellular gene to de-differentiate mammal somatic cells. Cellular degeneration of somatic cells can be induced by activating transcription of embryonic stem cell-related genes (e.g., oct3 / 4 ) and inhibiting somatic cell-specific and / or apoptosis-associated genes. Endogenous transcription systems can be modulated using synthetic transcription factors (activators and repressors), enabling faster and more efficient nuclear degeneration under conditions acceptable for clinical and commercial applications. The present invention further provides a cell obtained by such a method, a therapeutic method for using the cell in the treatment of a disease capable of treating stem cell therapy, and a kit for use in such a use.
