The present invention provides a recombinant E. Coli strain transformed into a gene having sequence number 1 as a hyperthermophilic archaeon-derived 1-Cys Prx mutant. The recombinant E. Coli strain is transformed into a recombinant expression vector containing a 1-Cys Prx mutant in which a part of a Prx gene of Thermococcus Kodakaraensis KOD1 is deleted, and thus can survive even when cultured under thermal stress conditions of 50°C or more and maximize a ROS concentration in cells. In addition, the ROS level in cells when culturing the recombinant E. Coli strain under high-temperature stress environments can be quantified and employed as a standard marker for ROS measurement of other cells. In particular, the present invention can measure the level of ROS that exists in excess in basal cell carcinoma, and thus can contribute to miniaturization of equipment for detecting cancer cells, and can be employed as an experimental model to evaluate efficacy of natural compounds and therapeutic agents for cancer.
