The present invention relates to a codonopsis extract with an increased lupenone content, which is able to be used for biotechnology, medicine, and food; and a manufacturing method thereof. The method includes: a first step 1) of manufacturing a codonopsis extract; a second step 2) of obtaining an ethyl acetate fraction extract by fractionating the codonopsis extract of the first step into water and ethyl acetate; and a third step 3) of manufacturing a methanol fraction extract by fractionating the ethyl acetate fraction extract of the second step into methanol and hexane. The codonopsis extract, of which lupenone content is six times higher than an existing case, is capable of solving problems of stability and side effects as an edible plant, and suppressing lipogenesis and cell differentiation in an obese animal model, thereby being applied to various purposes of food compositions or medical compositions for treating or preventing lipid-related metabolic diseases caused by fat accumulation.
