FIELD: biotechnology.SUBSTANCE: method of scanning gene mutations in human tumours is described. It includes amplification of analyzed gene fragment, melting duplexes of TaqMan probes with single-stranded amplicons, building peak melting, identification of mutations by the asymmetry of the melting peaks, featured by double-stranded scanning gene mutations by symmetric real-time PCR with two TaqMan probes used for post-amplification of DNA melting.EFFECT: method allows for the quantitative measurement of the number of copies of the analyzed gene in the original DNA sample, for simultaneous scanning of mutations in both amplicon threads, that improves reliability and sensitivity of the analysis, and provides additional information about the type of mutation.6 dwg, 1 tbl
