A lighting apparatus for evanescent wave fluorescence microscopy comprising: a light-transparent substrate (SB) having a refractive index greater than that of water; a light emitting device (EL) arranged inside said substrate, adapted to emit light radiation towards a surface (S) of the substrate, said light emitting device being arranged such that at least one portion said radiation reaches said surface with an angle of incidence greater than or equal to a total internal reflection limit angle for an interface between said substrate and water; and at least one opaque mask (MO), arranged inside or on the surface of said substrate so as to intercept a portion of said radiation which, in the absence of said mask, would reach said surface with an angle of incidence less than said angle limit. Application of such a lighting device to evanescent wave fluorescence microscopy.
