Methods for module synthesis of N-glycans and their arrays

The present disclosure relates to novel modular methods for making high N-type, hybrid and complex N-glycans. The present disclosure also relates to an exemplary array of synthesized N-glycans spotted on aluminum oxide coated slides. These arrays can be used to detect and analyze binding interactions between synthesized N-glycans and glycan binding molecules such as HIV-1 neutralizing antibodies. The present disclosure also relates to defining methods for identifying agents that bind to various types of molecules on the array, and defining structural elements of the molecules on the array that bind to those agents. The arrays and methods provided herein can be used to identify general epitopes, for drug discovery, and as analytical tools. The present disclosure also provides glycan and epitope determinants useful in the detection, diagnosis, repeat monitoring and prevention of pathological diseases such as HIV.
