Disclosed is a method of detecting a lipid bilayer formed in a cell of a nanopore based sequencing chip. The integrating capacitor is coupled to the lipid membrane. A lipid membrane is between the working and counter electrodes. An alternating current (AC) voltage is applied to the counter electrode. An analog to digital converter (ADC) periodically samples the voltage across the integrating capacitor. A change in response to the change in the AC voltage is determined at the voltage sampled across the integrating capacitor. Based on the change in response to the change in AC voltage, as determined at the sampled voltage across the integrating capacitor, it is detected whether the lipid membrane contains a lipid bilayer.
