PROBLEM TO BE SOLVED: To provide means which can obtain absolute determination data in an original sample from relative determination data obtained by a next-generation sequencer, and which can verify whether the relative abundance ratio after PCR obtained by the next-generation sequencer accurately reflects the abundance ratio of each microbial population in the original sample.SOLUTION: The invention relates to a nucleic acid consisting of a partial sequence which comprises at least one partial sequence represented in a capital letter in a nucleic acid consisting of a base sequence of a specific sequence, and which comprises, in both ends thereof, a part consisting of at least 15 or more of base sequences continuous from a partial sequence represented in a capital letter of a partial sequence represented in a small letter adjacent to the partial sequence represented in a capital letter; or a nucleic acid consisting of a complementary sequence thereof and a nucleic acid for using as a standard nucleic acid sample.
