Disclosed is a surface enhanced Raman spectroscopy detection method, comprising: dropping an extracted and purified sample detection liquid (4) onto a SERS substrate (1), so as to make the chemical molecules (41) of the detection sample liquid (4) be adsorbed on the SERS substrate (1); then dropping a highly volatile organic solvent (5) onto the SERS substrate (1) on which the chemical molecules (41) have been adsorbed, so as to make the chemical molecules (41) be redissolved in the organic solvent (5), thus making the surface of the SERS substrate (1) emerge; irradiating the SERS substrate (1) with light so as to volatilize the organic solvent (5) on the SERS substrate (1) by heating, leaving the chemical molecules (41) concentrated in a concentration region; irradiating the concentration region with laser light so as to make the chemical molecules in the concentration region be adsorbed deeper to the SERS substrate (1), thereby forming a spectral measurement region of a solid phase, and focussing the laser light on the spectral measurement region to carry out the Raman spectroscopy measurement.
