The present invention provides a bifunctional polypeptide having hypoglycemic and immunomodulatory effects and a use thereof, and belongs to the technical field of biochemical pharmacy. The bifunctional polypeptide is a fused polypeptide of a hypoglycemic polypeptide and an immunomodulatory polypeptide with or without a flexible linker peptide therebetween. The bifunctional polypeptide has functions of a GLP-1 analogue for promoting the proliferation of pancreatic islet β cells, reducing blood sugar and reducing glycated hemoglobin in patients with type 1 diabetes, and reducing blood sugar, inhibiting eating, and reducing glycated hemoglobin in patients with type 2 diabetes, and also has functions of a type 1 diabetes autoantigen immunoregulatory peptide for regulating immunity and inhibiting pathological immune attacks.
