The present invention relates to a method of preparing a mixed extract of Dendropanax morbifera, Hovenia dulcis, Coriolus versicolor, Artemisia capillaris, and Oenanthe javanica using response surface methodology. More specifically, optimal extraction conditions for the mixed extract, which bring an increase in extraction yield, DPPH radical scavenging activity, the total polyphenolic content, and ROS formation suppression activity, have been established through response surface methodology (RSM) in terms of extraction temperature, extraction time, and ratio of an extraction solvent to a sample. Therefore, the efficient production of the mixed extract of Dendropanax morbifera, Hovenia dulcis, Coriolus versicolor, Artemisia capillaris, and Oenanthe javanica with increased extraction yield, DPPH radical scavenging activity, the total polyphenolic content, and ROS formation suppression activity is possible, and the method can be beneficially used in the production of drink compositions containing the mixed extract.
