The present invention relates to a cross-linked extracellular matrix and a manufacturing method thereof, wherein the extracellular matrix is derived from a fibroblast cell line and is cross-linked to adjust physical properties. In addition, provided is a method for culturing human embryonic stem cells by using the cross-linked extracellular matrix. The extracellular matrix manufactured according to the present invention can have improved strength compared to a matrix which is not cross-linked and induce cell proliferation while maintaining an undifferentiated state when human embryonic stem cells are cultured. In addition, a period during which human embryonic stem cells can be cultured is remarkably increased by using the same matrix, and thus it is possible to reduce costs and a time required for cell culturing. Therefore, the cross-linked extracellular matrix derived from fibroblasts produced according to the method of the present invention can be effectively used for culturing human embryonic stem cells.;COPYRIGHT KIPO 2019
