The present invention relates to a method of decellularizing small intestinal submucosa and cell-free intestinal mucosa prepared therefrom. Provided are the decellularization method of small intestinal submucosa, comprising: (a) an enzyme treatment step of collecting small intestinal submucosa of mammals other than humans and treating an enzyme agent; (b) a freezing step of freezing the small intestinal submucosa treated with the enzyme agent; (c) a thawing step of thawing the frozen small intestinal submucosa; and (d) a centrifugal separation step of centrifuging the thawed small intestinal submucosa to selectively take an extracellular matrix part, and cell-free intestinal mucosa prepared therefrom.
