The present invention relates to a regeneration method using somatic embryogenesis of Prunus serrulata var. pubescens mature tree, which comprises the steps of: (a) inducing embryogenic cells by inoculating a plant root fragment in vitro derived from the Prunus serrulata var. pubescens mature tree into embryogenic cell induction media; (b) inoculating the induced embryogenic cells into a somatic embryo induction media to induce a somatic embryo; (c) inoculating the induced somatic embryo into somatic embryo-germinating media to induce a plantlet in vitro; and (d) purifying the induced plantlet in vitro.
