The present invention relates to a method for preparing a pellet of chondrocytes using a 3D cell co-culturing and differentiating plate, which is capable of increasing an expression ratio of chondrogenic differentiation. According to the present invention, the method comprises: a step (a) of proliferative- or sub-culturing single cell type induced pluripotent stem cells on a nanopattern sheet having a groove- or post-shaped pattern to prepare undifferentiated induced pluripotent stem cells; a step (b) of culturing the undifferentiated induced pluripotent stem cells in a 3D cell co-culturing and differentiating plate to form an embryoid body (EB) of the induced pluripotent stem cells; and a step (c) of seeding a single cell of the EB in a well of the 3D cell co-culturing and differentiating plate, adding a chondrogenic medium, and inducing chondrogenic differentiation in an incubator at 35 to 39°C to acquire a pellet of chondrocytes.
