摘要

The present invention relates to a vector for optimizing expression of Upc2p, which is a mutated transcriptional regulatory factor, and a sterol precursor overproduction method using the same. More specifically, expression of the mutated transcriptional regulatory factor (Upc2p) with reduced affinity to ergosterol is optimized to smoothen feedback adjustment such that an entire ergosterol biosynthesis path is amplified, thereby producing a recombinant yeast with increased biosynthesis performance for a sterol precursor, such as epoxy squalene and squalene, and a ginsenoside precursor such as dammarenediol. Accordingly, recombinant yeast strains produced by using a mutated UPC2 expression cassette and a substitution cassette using a CRISPR system can be used for production of recombinant yeast for massively biosynthesizing ginsenoside or ursodeoxycholic acid (UDCA) precursor materials through additional metabolic engineering. According to the present invention, a recombined expression vector includes a UPC2 mutated gene with a mutated ligand coupling domain portion of Upc2p derived from Saccharomyces cerevisiae.;COPYRIGHT KIPO 2019
