The present invention relates to an antithrombotic composition comprising a hexene fraction, an ethyl acetate fraction or a mixture thereof of a Diospyros kaki leaf ethanol extract as an active ingredient. More specifically, the present invention relates to a pharmaceutical composition and health functional food for prevention or treatment/amelioration of thrombosis through a blood coagulation inhibition activity and a platelet aggregation inhibition activity comprising, as an active ingredient, a fat-soluble hexene fraction, an ethyl acetate fraction or a mixture thereof obtained by sequentially organic solvent-fractioning an ethanol extract of dried Diospyros kaki leaves. As being proved through an embodiment of the present specification, a hexene fraction, an ethyl acetate fraction or a mixture thereof of a Diospyros kaki leaf ethanol extract as an active ingredient of a pharmaceutical composition and health functional food for prevention or treatment/amelioration of thrombosis according to the present invention, exhibit strong antithrombotic activity by the blood coagulation inhibition activity and the platelet aggregation inhibition activity, do not show hemolytic activity to human red blood cells at all, have excellent thermal stability, and do not exhibit loss in effects of inhibiting thrombogenesis-related enzymes and blood coagulation factors even at an acidic condition of pH 2 and within blood plasma. Accordingly, the present invention is extremely useful in pharmaceutical and food industries since the hexene fraction, the ethyl acetate fraction or the mixture thereof of the Diospyros kaki leaf ethanol extract is expected to be used for prevention and treatment of thrombosis such as ischemic stroke and hemorrhagic stroke through improvement of blood circulation, and the active ingredient is processed into various forms such as extract, powder, pill, tablet and the like to be taken at any time.
