The present invention relates to a recombinant antigen derived from Zika virus E protein and uses thereof. Specifically, provided are polynucleotides encoding Zika virus E protein domain III three times repetitively, a recombinant plasmid vector comprising the same, and a DNA vaccine composition capable of inducing an immune response against Zika virus by effectively expressing a Zika virus antigen protein. The present invention also provides a neutralizing antibody against Zika virus obtained by using the polynucleotides and a method for producing the same.
