The present invention relates to a tissue dyeing compound comprising synthesis complexes of dye and hyperbranched polymer as active ingredients, and a preparation method thereof. More specifically, a synthetic complex is prepared by using Rhodamine or Rose Bengal as a dye, and using a compound represented by chemical formula (1) as a hyperbranched polymer. During laparoscopic surgery for minimally invasive surgery and surgery with an endoscope or robot, even if stained for a long time, the synthetic complex is not smudged or erased. Therefore, there is no need for additional dyeing even if the operation time is prolonged, and the precise cell location can be confirmed while securing the safety interface at the time of surgery.
