The present invention relates to the use of anti-α-synuclein antibodies for diagnosing elevated levels of α-synuclein in the brain. In particular, the present invention relates to a method of assessing the level of α-synuclein in plasma or CSF following administration of an anti-α-synuclein antibody or antigen-binding fragment thereof to a test subject, wherein the antibody comprises blood in the brain. Or, to CSF in the brain, can bind to α-synuclein with sufficient activity to alter the net outflow of α-synuclein.
