To provide a method for generating a random gene plasmid library, a method for producing a transformant using the same, and a kit for use in the method, which are capable of simply introducing random gene sequence libraries into a plasmid by using one round PCR.SOLUTION: A template is subjected to a one round PCR by PCR amplification (polymerase chain reaction) of a random gene library primer and an auxiliary primer. The random gene library primer has a site in which the 5' end is complementary to the auxiliary primer, the 3' end is complementary to the template, and a random gene library is included between the 5' end and the 3' end. The auxiliary primer is complementary to the random gene library primer at the 5' end, is complementary to the template at the 3' end, and has no random gene part.SELECTED DRAWING: Figure 1
