The present invention relates to an anticancer composition comprising a recombinant adenovirus expressing an extracellular matrix degrading factor. The recombinant adenovirus according to the present invention significantly reduces collagen I, collagen III, pibronectin, elastin, etc., which are main components of the extracellular matrix in tumor tissue, and selects a therapeutic gene only in tumor cells by the proliferation of the virus. Highly expressed, exhibit an excellent antitumor effect. In particular, when the recombinant adenovirus is co-administered with an existing anticancer agent or immune checkpoint inhibitor, it has an existing anticancer effect, but is present in the tumor tissue of the co-administered therapeutic substance. Thus, the present invention could be utilized as a core technology in the field of cancer treatment, because the diffusion and distribution in the cancer can be significantly increased and the antitumor effect can be further improved.
