Use of a biological RNA scaffold with in vitro selection to generate a robust small molecule binding aptamer for a genetically codeable biosensor
Libraries of scaffolds from riboswitches and small ribozymes and methods of using them are provided herein. The scaffolds of the present invention provide aptamers that are easily identified and characterized by structural scaffolds. The nature of the scaffold is that these RNAs for binding to the reading domain facilitate manipulation of biosensors that function in vitro and in vivo. Also provided are biosensors, synthetic RNA and DNA agents, and methods of using them.
