The present invention discloses a method for collectively quantifying a target protein using next-generation sequencing. The method of the present invention comprises the steps of: (a) subjecting a sample to be analyzed to a target protein population present in the sample and treating the aptamer library to form a complex between each target protein and each aptamer that specifically binds thereto; Thereby forming a complex population of the target protein and the aptamer; (b) separating the complex population from unbound aptamers; and (c) changing the sequence of each aptamer in the complex population.
