METHOD OF EX VIVO PRETREATMENT OF HAEMATOPOETIC STEM CELLS AND PHARMACEUTICAL COMPOSITION FOR ENHANCING ENGRAFTMENT OF HAEMATOPOETIC STEM CELLS

The present invention is directed at a method of an ex-vivo pretreatment of haematopoetic stem cells (HSCs) for enhancing engraftment of haematopoetic stem cells, the method comprising the steps of obtaining a sample containing haematopoetic stem cells, admixing said sample with at least one prostacyclin analogue selected from the group consisting of treprostinil, iloprost, cicaprost or beraprost or pharmaceutically acceptable salts thereof together with forskolin, wherein the ratio of prostacyclin analogue and forskolin is 1:3, and incubating said mixture for a period of time sufficient to stimulate G alpha_s-signalling in said cells. The invention is further directed at a pharmaceutical composition for enhancing engraftment of HSCs after transplantation in patients suffering from bone marrow disease, the composition comprising at least one prostacyclin analogue selected from the group consisting of treprostinil, iloprost, cicaprost or beraprost or pharmaceutically acceptable salts thereof together with forskolin and stimulated haematopoetic stem cells obtained by the provided method. The invention is also directed at the use of the provided compostion for treating individuals suffering from bone marrow disease.
