This document presents libraries of cores obtained from riboswitches and small ribozymes, and methods for their use. The skeletons of this invention provide aptamers that are easily identified and characterized by the indicated structural skeleton. The nature of the backbone predisposes these RNAs to connect with reading domains to construct biosensors that function in vitro and in vivo. Biosensors, synthetic RNA agents, and synthetic DNA agents and methods for their use are also provided.
