The invention relates to methods and substances for detecting thiopurine methyltransferase polymorphisms (TPMT). A method for detecting TPMT polymorphisms comprises the following sequential steps: (i) providing a biological sample containing patient DNA; (ii) providing a first mixture comprising primers and two hydrolysis probes adapted for DNA target sequence amplification and TPMT gene polymorphism rs1800462 (TPMT * 2); (iii) providing a second mixture comprising primers and two hydrolysis probes adapted for DNA target sequence amplification and TPMT gene polymorphism rs1800460 (TPMT * 3B) detection; (iv) providing a third mixture comprising primers and two hydrolysis probes adapted for DNA target sequence amplification and TPMT gene polymorphism rs1142345 (TPMT * 3C); (v) providing a polymerase chain reaction (PCR) premix; (vi) adding the DNA and premix isolated from the biological sample to the first, second and third mixtures for quantitative PCR and amplification of the target nucleic acid; (vii) measurement of fluorescence and determination of TPMT gene polymorphisms by comparing amplification results with previously known sample values.
