CD33-specific chimeric antigen receptor (CAR) having a polypeptide structure selected from V1 and V3 as illustrated in Figure 2, wherein said V1 structure comprises (a) an extracellular ligand-binding domain comprising VH and VL of a monoclonal anti-CD33 antibody, (b) an FcγRIIIα hinge, (c) a CD8α transmembrane domain, and (d) a cytoplasmic domain that includes a CD3 zeta signaling domain and a 4-1BB costimulatory domain; and wherein said V3 structure comprises (a) an extracellular ligand binding domain comprising VH and VL of a monoclonal anti-CD33 antibody, (b) a CD8α hinge, (c) a CD8α transmembrane domain, and (d) a cytoplasmic domain including a CD3 zeta signaling domain and a 4-1BB costimulatory domain.
