Provided is an alcohol dehydrogenase mutant and application thereof in coenzyme regeneration, belonging to the technical fields of enzyme engineering and biological engineering. The alcohol dehydrogenase mutant is obtained by means of mutating the starting amino-acid sequence of 84 valine and/or 127 tyrosine of the alcohol dehydrogenase shown in SEQ ID No. 1; the alcohol dehydrogenase mutant has high activity on various alcohol co-substrates, and can catalyze such alcohol co-substrates to regenerate the coenzyme NADPH; in comparison with the wild-type alcohol dehydrogenase KpADH, the activity and catalytic efficiency of the present invention are higher; the kcat value for the co-substrate 1,4-butanediol may be up to 75.9 min-1, the value of kcat/Km may be up to 2009 min-1·M-1, and the Km value may be as low as 11.3 mM; the alcohol dehydrogenase mutant has higher value for industrial applications.
